Treatment
Allocation to intervention
Randomised controlled trial
Procedure for enrolling a subject and allocating the treatment (allocation concealment procedures)
A randomisation schedule will be provided from the unblinded biostatistician to the unblinded pharmacy staff in a password protected file which will be used solely by the unblinded pharmacy staff or treatment assignment. Based on the randomisation schedule the unblinded pharmacy staff will prepare code break envelopes containing the treatment assignment information. If needed for intentional unblinding for safety reasons, the code break envelope will be used in the event of an emergency if the pharmacy staff are not available.
Methods used to generate the sequence in which subjects will be randomised (sequence generation)
Part 1: Treatment is randomised 1:1 (BMB-105: Placebo) in the Sentinel group, and 4:1 (BMB-105: Placebo) for each of the SAD cohorts.
Part 2: Treatment is randomised 1:1 (BMB-105 in fed / fasted state) crossed over.
Part 3: Treatment is randomised 4:1 (BMB-105: Placebo) for each cohort.
The randomisation is a computer generated random sequence.
